Barclay, one of Scotland’s outstanding performers in the 2010 RBS 6 Nations, will claim the armband back from full-back Bernardo Stortoni and is relishing the prospect of leading the team out at the Firhill Stadium.
"I captained the team a couple of times last year and enjoyed the challenge but we've got guys around me with experience and who have captained the team in the past, so they will be helping me out," said Barclay.
“There are some guys in there who have a lot of talent and maybe just need that little bit of leadership and a push in the right direction.
“The players have all the attributes they need and they will play well, but they need to channel that into a performance on a weekly basis.”
The Warriors were defeated 23-11 by Newport Gwent Dragons after opening the season with victory over Leinster and Barclay admitted his frustration from watching from the sidelines.
"It was difficult watching the Leinster game," he said.
"Obviously you want to be involved, but when the guys won I was delighted, especially with the manner in which they won.
“Watching the Dragons game last weekend was a different kind of frustration.
"But I'm certainly looking forward to being back, being involved and being in a Glasgow Warriors shirt again.
“It’s a long season and I was thinking it’s maybe not a bad thing to have a break but you miss it straight away and I just wanted to get back out there.”
